| I. Background | |
| 细胞因子胸腺基质淋巴生成素(TSLP)是一种 I 型细胞因子，是Ⅱ型免疫应答的关键介 质和 T 辅助 2 (TH2)细胞介导的疾病的启动子，包括哮喘和特应性皮炎(AD)。它顺利获得激活 抗原呈递细胞在 T 细胞群的成熟中发挥重要作用。多种触发因素可诱导 TSLP 的产生，包 括损伤、TLR2 (toll 样受体 2)、TLR3 和 NOD2 的配体、蠕虫、细菌和病毒感染、蛋白酶 (如胰蛋白酶和木瓜蛋白酶)和促炎细胞因子。 TSLP 结合由 TSLPR(胸腺基质淋巴生成素受体)和 IL-7 受体α链(IL-7Rα)组成的异二聚 体受体，启动由 JAK1 和 JAK2 介导的级联磷酸化。随后 STAT5A 和 STAT5B 的募集和激活 最终导致 Th2 相关细胞因子 IL-4、IL-5、IL-9 和 IL-13 的转录和产生。 |
