| I. Background | |

4-1BB [也称为 CD137 或 TNF 受体超家族成员 9 (TNFRSF9)] ，是糖基化的 I 型膜蛋白，属于 TNFR 超家族成员，由四个胞外富含半胱氨酸的假重复结构域组成。4-1BB 的胞质区包含 TNF 受体相关因子 (TRAF) 结合基序。4-1BB 表达主要发现于活化的细胞毒性CD8 + T 细胞和辅助性 CD4 + T 细胞的表面，活化后，NK 细胞、B 细胞、单核细胞和树突状细胞中可诱导 4-1BB 表达。作为可诱导的共刺激分子，4-1BB 可增强 T 细胞对抗原的反应。在 T 细胞上被 4-1BBL 上调和三聚化后，4-1BB 会募集 TRAF 衔接蛋白至细胞浆TRAF 结合基序，从而启动共刺激信号传导。
4-1BBL (TNFSF9) 是 4-1BB 的主要配体，主要在 APC 、B 细胞、巨噬细胞和其他细胞类型中表达，并与 4-1BB 结合，顺利获得 TRAF1 和 TRAF2 诱导信号传导，从而激活 NF-κB、AKT、p38 MAPK 和 ERK 通路。NF-κB 诱导 BCL-XL、BCL-2 和 c-FLIP 的转录，并刺激IL-2、IL-4 和 IFN-γ 的表达，从而促进 T 细胞的存活和细胞毒性活性。因此，4-1BB/4- 1BBL 信号通路可以促进不同类型的细胞存活。
