| I. Background | |
| TM4SF1 基因编码的跨膜蛋白 4 超家族成员 1（TM4SF1）是一种四次跨膜糖蛋白，属 于 Tetraspanin 超家族，在多种肿瘤中异常高表达。该蛋白顺利获得四个疏水跨膜结构域与整合 素、受体酪氨酸激酶等分子相互作用，参与细胞信号转导、黏附和迁移，促进肿瘤血管生 成、上皮-间充质转化（EMT）及肿瘤干细胞自我更新，显著增强癌细胞侵袭和转移能力。 临床研究表明，TM4SF1 在肺癌、乳腺癌、肝癌、结直肠癌及卵巢癌等多种恶性肿瘤 中显著上调，其表达水平与肿瘤转移及不良预后密切相关。研究发现，TM4SF1 顺利获得激活 JAK2-STAT3 信号通路，促进乳腺癌向肺、骨、脑等远端器官转移；同时，它还参与肿瘤 耐药机制，降低治疗效果。 |
